Involvment of IL-1 beta Mediated NF-Kappa B Signalling Payhways to Downregulate Prostate Specific Antigen And Cell Proliferation in LNCaP Prostate Cancer Cells.

Background: The aim of the present work was to study the involvement of nuclear factor kappa B mediated by Interleukin-1 beta on cell proliferation and Prostate Specific Antigen production of LNCaP prostate cell lines and the possible cross talk with AKT signalling pathway. Methods: Nuclear factor kappa B and AKT were performed by Western Blot analysis from LNCaP cells treated by Interleukin beta. Subsequently, LNCaP cell proliferation and Prostate Specific Antigen production were measured. Results: The treatment of LNCaP cells with Interleukin1 beta inhibited cell proliferation and decreased PSA production. AKT pathway was not sensitive; whereas the phosphorylation of nuclear kappa B was followed by the cytokine's effect. The Prostate Specific Antigen production and the proliferation of LNCaP cells were down regulated by nuclear kappa B mediated by Interleukin1 beta promoting anti-apoptotic signaling and co-suppressor factors of Prostate Specific Antigen expression. Conclusion: Interleukin1 beta through nuclear kappa B activation provides a rationale for therapeutic approaches in the anticancer treatment of prostate.
