Hypolipidemic and anti-atherogenic activity of aqueous extract of Gleditsia sinensis Lam. in rabbits fed a high-lipids diet.

The efficacy of hypolipidemic and anti-atherogenic in Gleditsia sinensis Lam. fruit aqueous extract (GAE) was investigated in Japanese white rabbits fed on high-lipids diet. Rabbits were divided into four groups: the first two groups of rabbits received normal and high-lipids diet, respectively, and the remaining two groups received high-lipids diet supplemented with GAE (12 and 6mg/kg B.W.). The groups with high-lipids diet were administered i.v. with bovine serum albumin (BSA) in the 4th week for atherosclerosis modeling. The concentrations of triglycerides (TGs), total cholesterol (TC) and low density lipoprotein cholesterol (LDL-C) were determined in rabbits at the start of the experiment, in 4th, 8th weeks and at the end of the study (14th week). And the activities of aspartate aminotransferase (AST), alanine aminotransferase (ALT) and creatine kinase (CK) were determined simultaneously. The rabbits were sacrificed and the aorta and liver were removed at the end of the experiment. Atherosclerotic plaques were assessed by oil-red and H&E staining, while the lipids level (TG, TC, LDL-C) in tissue was determined in aorta and liver homogenate. Results showed that GAE could significantly decrease TC and LDL-C levels and retard aortic atherosclerosis. Hence GAE can effectively prevent the progress of atherosclerosis. This is likely due to hypolipidemic effect of GAE.
