OBJECTIVE: To investigate the effect of acute blood glucose fluctuation on the function of rat pancreatic β-cells in vivo. METHODS: In this work, we reported a novel method to induce animal models of acute blood glucose fluctuation and persistent hyperglycemia by regularly intermittent and persistent intravenous injecting high concentration glucose solution. Both malondialdehyde (MDA) and superoxide dismutase (SOD) activity in pancreatic islets were examined by colorimetric kits. Protein expression was tested by Western blot. Pancreatic β-cell apoptosis was examined by immunofluorescence. RESULTS: The level of plasma insulin was significantly lower in persistent hyperglycemia group and blood glucose fluctuation group than in control group during 48h (P<0.05). The sensitive index of plasma insulin in blood glucose fluctuation group was the lowest among three groups (P<0.05), and the ratio of MDA/SOD in persistent hyperglycemia group was the highest among three groups (P<0.05). Meanwhile, the number of apoptotic β-cells in the pancreatic tissue significantly increased in persistent hyperglycemia group and blood glucose fluctuation group (P<0.05), and it was significantly higher in blood glucose fluctuation group than in persistent hyperglycemia group (P<0.05). CONCLUSION: The damage to pancreatic issue may be associated with the increased level of oxidative stress caused by acute blood glucose fluctuation. |
