Several novel oral anticoagulants have been approved by the U.S. Food and Drug Administration and the European Medicines Agency for treatment of specific thrombotic conditions. The direct thrombin inhibitor, dabigatran, and direct factor Xa inhibitor rivaroxaban have certain advantages over warfarin, an oral vitamin K antagonist that has been used for over 50 years. Among these advantages are fewer drug-drug and drug-food interactions, predictable pharmacokinetic profiles allowing fixed dosing once or twice daily, and most important, avoidance of the need for routine coagulation monitoring. © 2012 International Society on Thrombosis and Haemostasis.
