Changes in expression profiles of antioxidant enzymes in diabetic rat kidneys.

BACKGROUND: In diabetes mellitus, increased formation of reactive oxygen species (ROS) due to high level of glucose in both blood plasma and tissues creates oxidative stress, and damages the tissues. Antioxidants together with the antioxidant enzymes are very important in order to protect the cells against oxidative damage. METHODS: Differential expressions of both mRNA and proteins of major antioxidant enzymes in streptozotocin induced diabetic rat kidneys were measured with the help of real time PCR and western blot analysis respectively. Furthermore, effects of two strong antioxidants α-lipoic acid (LA), vitamin C (VC) and their combination on the regulation of both expressions and the activities of antioxidant enzymes were also studied. RESULTS: Though mRNA expressions were greatly induced, both catalase (CAT) and glutathione peroxidase (GPx) activities were found to be reduced in diabetic rat kidney tissues when compared to controls. Even though, no significant change was observed in superoxide dismutase (SOD) activities, the decrease in SOD mRNA and protein expressions reflected a transcriptional repression in diabetic animals. Individual LA application increased CAT activities toward the control values though suppression was observed in mRNA expressions. Combined administration of LA and VC increased the activities of both CAT and SOD demonstrating a post-translational effect. GSH concentration showed a significant decrease in diabetic kidney tissues and LA treatment increased the GSH level toward the control levels. CONCLUSIONS: All data showed the importance of post-transcriptional and translational regulation of the antioxidant enzyme activities against oxidative stress which is associated with diabetes.
